Effect of Trifluoperazine amd Isoquinolinesulfonamide in the Cat Gallbladder Muscle Tone

Abstract
|

|
Transversely oriented cat gall bladder rings(2mrn wide) tested in vitro maintained basal tone spontaneously. Cholecystokinin octapeptide(OP-CCK), acetylcholine(ACh) and potassium chicride(KCl) contracted the muscle in a concentration dependent manner.
1-(5-Isoquinolinesulfopyl)-2-methylpiperazine dihydroOhloride(H-7), a putative protein kinase C(PKC) inhibitor, significantly reduced basal tone, but did not affect OP-CCK-, ACh- or KC1-induced contraction.
Trifluoperazine(TFP), a putative calmodulin inhibitor, did not affect the basal tone but significantly reduced OP-CCK-, ACh- and KC1- induced contractions.
The result suggests that there are at least two different pathways to obtain net gallbladder muscle tension. One is H-7 sensitive, i.e., protein kinase C dependent, and contributes to the maintainance of the basal tone. The other is TFP sensitive, i.e., calmodulin dependent, and contributes to the development of tension induced by stimulators such as OP-CCK, ACh or KC1.(Author)
